Ellsinore is located at 36°56′4″N 90°44′47″W / 36.93444°N 90.74639°W (36.934514, -90.746277)[3].
According to the United States Census Bureau, the city has a total area of 0.4 square miles (1.0 km2), all of it land.

As of the census[1] of 2000, there were 363 people, 162 households, and 99 families residing in the city. The population density was 830.7 people per square mile (318.5/km²). There were 184 housing units at an average density of 421.1 per square mile (161.5/km²). The racial makeup of the city was 98.07% White, 0.55% Native American, and 1.38% from two or more races. Hispanic or Latino of any race were 0.28% of the population.
There were 162 households out of which 25.3% had children under the age of 18 living with them, 47.5% were married couples living together, 10.5% had a female householder with no husband present, and 38.3% were non-families. 36.4% of all households were made up of individuals and 25.3% had someone living alone who was 65 years of age or older. The average household size was 2.20 and the average family size was 2.85.
In the city the population was spread out with 22.6% under the age of 18, 9.1% from 18 to 24, 21.2% from 25 to 44, 20.4% from 45 to 64, and 26.7% who were 65 years of age or older. The median age was 43 years. For every 100 females there were 78.8 males. For every 100 females age 18 and over, there were 75.6 males.
The median income for a household in the city was $17,143, and the median income for a family was $27,188. Males had a median income of $26,125 versus $16,250 for females. The per capita income for the city was $11,444. About 10.6% of families and 19.7% of the population were below the poverty line, including 22.8% of those under age 18 and 20.8% of those age 65 or over.

On Wednesday, December 19, 2007, Ellsinore Mayor William (Bill) Fulford was killed in an automobile accident. The vacancy has been filled by David Bowman.[4]
Incumbent Mayor David Bowman beat challenger Yvonne Collins by a landslide in the 2008 Mayoral race on April 8, 2008, capturing 74% of the vote.
Carl "Andy" Bowman, Brenda (Hahn) McGehee, and Ben Theis, all challengers, were elected to the East Carter County R-II School Board on April 8, 2008.

At approximately 8:50 PM central time on the night of April 6, 2009, a Cessna 172 airplane that was stolen from a flight training school in Ontario, Canada landed on U.S. Highway 60 in Ellsinore, flown by a despondent flight student wishing to commit suicide. He safely landed and hitched a ride to Simmons' Convenience Store, where he was seen by employees, who alerted law enforcement officials, who promptly arrested him and took him to the Butler County jail in Poplar Bluff.
