State Route 281 (SR 281) is an east–west state highway in the northwestern portion of the U.S. state of Ohio. Its western terminus is at U.S. Route 24 approximately 4 miles (6.4 km) northeast of Defiance, and its eastern terminus is at U.S. Route 6 nearly 1½ miles north of Bradner. The majority of the route is known as the Defiance Pike.

- 1935 – Extended west to State Route 18 and east to State Route 25 along previously unnumbered roads[2].
- 1939 – Extended east to its current terminus via a previously unnumbered road from State Route 25 to Wayne and via a former alignment of State Route 297 from Wayne to 1½ miles north of Bradner[2].
- 1972 – Extended to its current western terminus along with State Route 18 for just over 1½ miles west, and then north on a newly constructed road[2].
