State Road 53 is a 20-mile-long north–south route in northern Florida. Its southern terminus is an interchange with Interstate 10 (SR 8) near Lee and Madison; the northern terminus is at the Georgia state boundary near Cherry Lake, Florida. The southernmost six miles (10 km) of SR 53 is also signed State Road 14 Truck, a rare bannered State Road.
State Road 53 continues northward from the state line as State Route 333; a southbound continuation of SR 53 is Madison County Road 53 and Lafayette County Road 53, which ultimately ends at an intersection with SR 51 north of Mayo.
