U.S. Route 378 (US 378) is a spur of US 78. It runs for 234 miles (377 km) from Conway, South Carolina at US 501 to Washington, Georgia at US 78.

US 378 passes through the states of South Carolina and Georgia.

In South Carolina, US 378 provides an important link between Columbia and the cities of Sumter, Lexington, and Saluda.

US 378 and US 1 run concurrent in Lexington for a short while before diverging.

In West Columbia, US 378 runs concurrent with US 1 again. The two roads cross over the Congaree River and enter Columbia.

In Columbia, US 378 runs concurrent with US 76 for approximately 40 miles (64 km) until they diverge in Sumter.
