Caroline Wozniacki was the defending champion, but she lost to Maria Sharapova in the third round.

Simona Halep, Naomi Osaka, Petra Kvitová, Elina Svitolina and Karolína Plíšková are in contention for the WTA no. 1 singles ranking at the middle of the fourth round. In order to have a chance at taking over the top ranking from Halep, Osaka and Kvitová must reach at least the semifinals, while Svitolina and Plíšková must reach the final. Eleven players had a theoretical chance of becoming no. 1 at the start of the tournament (ten if Halep played her first match).[1] Sloane Stephens, Angelique Kerber, Wozniacki, Kiki Bertens, Aryna Sabalenka and Daria Kasatkina were eliminated from contention as the tournament progressed.

Serena Williams will attempt to equal Margaret Court's all time record of 24 Grand Slam singles titles.

Amanda Anisimova became the first player born after 2000 to advance to the fourth round of a Grand Slam singles tournament.

This was the first Australian Open to feature a final set tiebreak. When the match reaches 6–6, the first player to reach 10 points and be leading by at least 2 points wins the match. Katie Boulter and Ekaterina Makarova became the first players to contest this tiebreak in the main draw, with Boulter emerging victorious.
