State Route 372 begins with its intersection with SR 140 in the western part of Alpharetta, Georgia and travels to the north, through unincorporated, but rapidly growing, areas of Fulton and Cherokee counties.

After intersecting with SR 20 and SR 369 in Free Home, the route turns more towards the northwest, crossing the Etowah River, then entering Ball Ground. In Ball Ground, the route begins a concurrency with SR 5 Business. In Nelson, the two routes utilize a short expressway stub before the routes end at SR 5 and SR 515.
