Talk:Tsix
Biol 460 Project
We are thinking of breaking the page up into sections. We also added links to this page from Xist and Dosage Compensation. — Preceding unsigned comment added by Iamwillthinnes (talk • contribs) 03:35, 19 February 2014 (UTC)
We have covered all of the suggestions. Future planned improvements include creating a graphic for the page and adding Tsix links to related pages, both per Dr. Ogilvie's suggestion. Iamwillthinnes (talk) 16:18, 15 April 2014 (UTC)
Comments from User: Emileemagnusen:
- I think the "Function in mammals" section is good, but I think it might be too jargon heavy. I would try to explain the same concepts just in plain language. The average reader may not be familiar with all of the terms you are referring to.
- I clarified dosage compensation and the need for X inactivation. I also added several links here and throughout the article wherever there were scientific terms.Jtrivedi92 (talk) 02:52, 15 April 2014 (UTC)
- Add links to other Wikipedia articles within your content.
- I added links to several relevant scientific terms throughout the article, such as imprinting, dosage compensation etc.Jtrivedi92 (talk) 02:52, 15 April 2014 (UTC)
- Add more content to "Effect of Tsix mutations." It may be interesting to read about mutations that result in abnormal X and Y chromosome pairings (XXY, etc.)
- Agreed. However we haven't been able to find any articles discussing specific Tsix mutations in humans.Iamwillthinnes (talk) 14:49, 15 April 2014 (UTC)
- It might be helpful to add a subsection within the "Function in mammals" section that goes into more detail about imprinting and how it relates to the Tsix gene. Or, you could just add a link to another Wikipedia article on imprinting. Either way, readers should have some reference as to what imprinting is.
- added link to imprinting article, and a brief descriptionJtrivedi92 (talk) 02:52, 15 April 2014 (UTC)
- "Regulation" section is also very science language heavy.
- Fixed to make it more readable. Iamwillthinnes (talk) 15:31, 15 April 2014 (UTC)
- This is just a suggestion, but you could add a section about the history of the Tsix gene. Who discovered it? When?
- We know the first group to identify the gene, but I don't know if this is necessary for this Wikipedia page. For one, the Xist gene page does not have a similar section. While I do like the idea, I don't think that it would add to the page.Iamwillthinnes (talk) 15:36, 15 April 2014 (UTC)
Comments from Estephe9
- Lead suggestions
- Euchromatic is a term not all readers (myself included) aren't familiar with. I recommend either defining this or linking it to an article about the term (perhaps euchromatin).
- added link to euchromatinJtrivedi92 (talk) 02:54, 15 April 2014 (UTC)
- The last 3 sentences of your lead need citations
- Added a link to the end of the lead.Iamwillthinnes (talk) 02:59, 15 April 2014 (UTC)
- This is a stylistic suggestion, but the wording of the second and third sentences is a bit confusing. You may want to have a link to the female karyotype, X-inactivation, or even a brief explanation of dosage control to help make sense of this a little more.
- "Xist is expressed from the X chromosome in females and serves to inactivate the other X chromosome." This makes it sound as though there is only one X chromosome in females, which is confusing because you then mention a second X chromosome.
- "Tsix prevents the accumulation of Xist on one female X chromosome to maintain the active euchromatic state of the chosen chromosome." Again, you may want to explain that, for dosage reasons, one X chromosome must remain active and the other repressed, which is why Tsix is necessary to prevent Xist from accumulating on one of the two X chromosomes.
- fixed this for clarity, added a more thorough description of dosage control and X inactivationJtrivedi92 (talk) 02:54, 15 April 2014 (UTC)
- Function in mice
- Add a link to [imprinting (genetics)]]
- Nit-picky, but you may want to add that Xist inactivates the paternal chromosome by condensing it and maybe link that to chromosome methylation or something like that to help better explain that process. — Preceding unsigned comment added by Estephe9 (talk • contribs) 15:46, 20 March 2014 (UTC)
- did this! and added link to histone methylationJtrivedi92 (talk) 02:54, 15 April 2014 (UTC)
- Effect of Tsix Mutations
- Nit-picky, but the first sentence was kind of long, which confused me.
- Reworded the first few sentences. Iamwillthinnes (talk) 16:16, 15 April 2014 (UTC)

- Regulation
- You may need a source for the last sentence of the first paragraph if it's not found in the source used to write the rest of that paragraph.
Estephe9 (talk) 04:22, 22 March 2014 (UTC)
- Got it, thanks. Iamwillthinnes (talk) 16:08, 15 April 2014 (UTC)
Comments from Akhan50699
- The first paragraph under "Functions in Mammals" needs a citation.
- Under "Regulation in cell differentiation" you might consider linking to Rex1 for the reader's reference.
- Source 7 also briefly mentions the X inactivation in bovine-it might be useful to include this information (and try to find information about the Tsix mutations in other animals if possible) under the "Effects of Tsix mutations" because this section only has information about mice so far.
Akhan50699 (talk) 03:16, 22 April 2014 (UTC)
- This is a little picky, but under the "function in mammals section," in sentence 3, the words "over accumulation" have no dash between them, while in sentence 4 they do have a dash between them. For the sake of consistency, you might change that so they appear the same in both sentences.
- Under "mutations," you might link out to null allele.
- For clarification of the first sentence under "Tsix in humans," when you say "imprinting does not occur" do you mean imprinting of Tsix?
- Under "Tsix in Humans" you might link out to vestige

Comments from Rraju2
- Adding the diseases or conditions associated with the dysfunction or mutation of this gene in humans (if relevant) would be interesting.
- A history section could also be beneficial to readers
- Possibly add more links to other articles

Gene summary box and flowchart
I added the gene summary box using a similar format to the one for the Xist gene. If this is incorrect, please feel free to change it. I also added a simplified flowchart of Tsix's role in Xist regulation, but I can't seem to make the image go to the side of the page. If anyone can correct this, please feel free to do so.Jtrivedi92 (talk) 19:27, 23 April 2014 (UTC)
Comments from Biolprof
- You may find some useful information that should be included on the OMIM page, but you may find this material too technical for wikipedia. I have only glanced at it, but if you haven’t already read through this and the associated sources, I think you will find updates on Tsix regulation and some other topics.
- Function in mammals section lacks references in the first portion.
- Function in mammals, last part: These sentences could also be edited to be less technical/formal and easier to understand.
- Edited to make less technical/ reworded for easier understanding. Iamwillthinnes (talk) 22:43, 10 May 2014 (UTC)
- Tsix in humans: a brief explaination of CpG island is needed in addition to the link.
- Added a link to epigenetic regulation. Iamwillthinnes (talk) 22:43, 10 May 2014 (UTC)
- reference needed for the statement “An autosome may be more likely…”
- Second to last sentence: delete “also.”
- Regulation in cell differentiation: Lower case in header except first word.
- Regulation in cell differentiation: Does this apply only to mouse? If so, move above human section. If not, make that clear in the text.
- Moved section. Iamwillthinnes (talk) 22:43, 10 May 2014 (UTC)
- Fix or add the wikilink to Tsix (gene) on other WP pages including: XIST (gene), Dosage compensation, Cis-natural antisense transcript. It is shown in red in several places because it has the incorrect page name. Biolprof (talk) 14:59, 25 April 2014 (UTC)
Comments from Biosthmors
Hello and thanks for your contributions. A few comments:
- Maybe we can make the WP:First sentence more accessible. I didn't know what an RNA gene was, so can we post that the portion is non-coding?
- Added it in, thanks for the comment. Iamwillthinnes (talk) 23:12, 8 May 2014 (UTC)
- In the WP:LEAD, what does "function as the antisense" mean? Are we trying to say that it always binds the other single strand of RNA to inactivate it? Maybe we can say so.
- I clarified this and made it easier to understand. Let me know if you think I should further clarify anything in the lead. Thanks, these are helpful. Iamwillthinnes (talk) 23:17, 8 May 2014 (UTC)
- I wonder if the sentence -- "Xist is expressed from one of the X chromosomes in females and serves to inactivate the other X chromosome" -- and some of the background information in the article is always needed. Maybe the sentence can be reworded to make Tsix the first word? I wonder if we are straying off topic a bit. When I write a Wikipedia article, I tend to focus on the topic exclusively, unless I write a specific background section. Maybe one would be valuable in this article. That way readers can get right to Tsix information (or the background), if they desire.
- What would be the best way to add a background section to this article? It certainly needs one. Understanding Tsix is incomplete without Xist and Xinactivation background. Iamwillthinnes (talk) 23:22, 8 May 2014 (UTC)
Hi Biosthmors - Iamwillthinnes requests your feedback. Iamwillthinnes, can you see how I notified both of you in this message? Biolprof (talk) 15:44, 9 May 2014 (UTC)
I hope this helps. Best. Biosthmors (talk) pls notify me (i.e. {{U}}) while signing a reply, thx 22:52, 2 May 2014 (UTC)
Comments from User:Ecapelle
- Are there any pictures you can use of Tsix or PRDM14 that are available through Wikimedia Commons?
- Under Tsix in Humans you use the phrase "evolutionary vestige". Could you define or explain this a little bit more?
- I suggest wikilinking some more phrases like: evolutionary vestige, Rex-1, pluripotency network, PRDM14, etc.
- Is Tsix present in other organisms or mammals? You go into detail about humans and a little about mice, but if it has evolutionary roots, is it still present?

Comments from User: Jfriend2
- work on expanding the mutation section and Tsix in humans to more than just one paragraph
- in the lead table, you can remove the sections for which information is not available which happens a couple of times
