Is this game notable enough to be included? That is, are there any published references to this game anywhere? At least www.pagat.com and US Playing Card Company rules collection do not mention this game. With a quick google search, I was able to find only one mention of the game. (Admittedly, using google is difficult in this case, since there exist too many other things named 'mille'.) Please see Wikipedia: No original research.

Second question: How do the rules about a meld of eight cards work? If you collect several such sets in one hand, can you get several marks during that hand? If you, in addition, do not use wilds anywhere, can you get one mark for a meld of eight cards, and another for not using wilds. It appears to me that Wikipedia: No original research has nothing to do with a card game. No opinions or ideas are given here, just rules to a card game. I'll admit that Mille isn't notable, as evident by your statement that the game isn't on Pagat.com or included in US Playing Card Company's rules collection. However, Mille is a card game that is slowly making gaining popularity, especially among poker players. To answer your questions on gameplay, you would receive one mark for each 8-card meld. It would difficult, but certainly worth it if it occurs. If you went out "natural" AND had an 8-card meld, it would be two marks as well. Both marks are independent of each other. If you had an 8-card meld and still lost that particular hand, you would also receive the mark. However, if you lost the game, you wouldn't receive anything for the mark.
