Premier Division
The Premier Division consisted of 22 clubs.

- AFC Kempston Rovers – promoted to Southern League Division One Central
- Northampton Spencer – club folded
The following 2 clubs joined the Premier Division before the season -
- Peterborough Sports – promoted from Division One
- Northampton ON Chenecks – promoted from Division One

Promotion criteria
To be promoted at the end of the season a team must:[1]
- Have applied to be considered for promotion by 30 November 2016
- Pass a ground grading examination by 31 March 2017
- Finish the season in a position higher than that of any other team also achieving criteria 1 and 2
- Finish the season in one of the top three positions

- Daventry Town – voluntarily relegated from Northern Premier League Division One South
- Melton Mowbray – promoted from Leicestershire Senior League Premier Division
- Whittlesey Athletic – promoted from Peterborough and District League Premier Division
