It has been suggested that Granzyme B cleavage of Bid is an artifact. Also, the model for granzymes entering APCs is outdated. Now people think that there is a two step model whereby perforin creates holes in the plasma membrane which allow an influx of calcium triggering membrane repair mechanisms. Then these repair mechanisms result in endocytosis of the plasma membrane which brings granzymes to early endosomes.
