Pour Plate Method : 1. Dillution of sample. a.take 9ml normal saline in 5 tubes and mark them as 1,2,3,4,5. and then take 1 ml of bacteria/sample and put it in tube 1(now the concentration of the 1st tube is 10ml ). 2.Now from 1st bube which is containing 10 ml of diluted sample take 1ml and put it in tube no. 2 3.Do this same in next tubes . 4.Now from all the 5 tubes inoculate the samples to madia plates.

{REDUCTION OF COLONYS = INCREASE IN DILLUTION FACTOR}

Formulla of counting bacteria= 30×D.F(dilution factor)/30×10000= no./ml bacteria.
