Her keel was laid down August 15, 1938, by AG Weser of Bremen. She was commissioned August 26, 1939 with Kapitänleutnant Wilhelm Ambrosius in command.

U-43 conducted fourteen patrols, as part of the 2. Unterseebootsflottille and 6. Unterseebootsflottille.

Fate

On July 30, 1943, U-43 was sunk in position 34°57′N 35°11′W / 34.950°N 35.183°W / 34.950; -35.183, by a Fido homing torpedo from an Avenger aircraft from the USS Santee.
