WDTC1
[edit]WDTC1 is also known as the "obesity" gene due to its connection and correlation with levels of adipose tissue. WDTC1 is a gene that codes for a protein acting as a suppressor in lipid accumulation. WDTC1 protein consists of seven WD40 domains, three transient receptor potential channel (TPR) protein-protein interaction domains, DDB1 binding elements, and a prenylated C-terminus.[1] Reduced expression or disruption of WDTC1 gene is associated with obesity, increased triglyceride accumulation, and adipogenesis. WDTC1 is a factor in a complex comprising of DDB1, CUL4, and ROC1 that restricts transcription in adipogenesis. [2]
Structure
[edit]Adipose (ADP) is a evolutionary conserved gene that codes for a protein containing WD40 domains, TPR domains, DDB1 binding elements, and a prenylated C-terminus. The ADP gene homolog in humans is the WDTC1 gene.
Phenotype
[edit]Expression of the gene in model organisms results in suppression of lipid accumulation and formation by restricted transcription in lipid formation. Studies of phenotype of mice showed that having a loss of an allele resulted in obesity, poor metabolic profiles, and excessive lipid storage. Transgenic expression of the WDTC1 gene in mice showed the opposite effect with mice having less adipose.[3]
