Flavia Pennetta is the defending champion, but chose to not participate after retiring from professional tennis at the end of the 2015 season, thus she can't defend her title.

Serena Williams will attempt to set a new Open Era record by winning a 23rd Grand Slam singles title. She currently shares the record of 22 with Steffi Graf. Williams will also attempt to win a record-breaking seventh US Open title in the Open Era. She currently shares the record of six titles with Chris Evert.

At the start of the tournament, the top four seeds (Williams, Angelique Kerber, Garbiñe Muguruza and Agnieszka Radwańska) are in contention for the world No. 1 ranking.
