Most of the county routes in Erie County, New York, act as primary roads in the less developed areas and also serve to interconnect the various villages and hamlets of the county. Municipalities served include the cities of Buffalo and Tonawanda, and the villages of Angola, Evans, Farnham, Lancaster, Springville, East Aurora and Depew. Routes 385 to 416 are listed below.

County Route 385 is an overall east–west route in the town of Wales. Named Schang Road, CR 385 begins at Strykersville Road (NY 78), west of Wales Hollow. The roadway travels north from NY 78 for 4/10th of a mile and travels east until an intersection with Woods Road, where the roadway turns and travels south for 7/10th of a mile until turning east again. CR 385 continues until the Wyoming County line where it ends as well as the road name, and Bloecher Road takes over into Wyoming County.

County Route 401, named Ward Road, is a short east–west route that lies on the Orchard ParkBoston town line, with Orchard Park to the southeast and Boston to the northeast. Ward Road itself begins at Boston Ridge Road (NY 277); however, the CR 401 designation begins 1.7 miles (2.7 km) to the east at an intersection with CR 371 (Scherff Road). From here, CR 401 travels east for only a half-mile to the roadway's end at CR 442 (Cole Road).

County Route 402, named Liberia Road, is an east–west route that lies entirely within the town of Marilla. It begins in the hamlet of Porterville at an intersection with Two Rod Road (unsigned NY 954G). From there, CR 402 travels east for 2.8 miles (4.5 km) to its end at a junction with CR 355 (East Blood Road). Former NY 358 overlapped CR 402 from its west end where it would follow eastbound Liberia Road to the intersection of Four Rod Road, where it would continue south, until it ended at US 20A in the town of Wales. The NY 358 designation was removed in April 1984.

CR 403, named Miller Road, begins at South Protection Road (CR 531) in the hamlet of Protection, located in the northeastern corner of Sardinia near NY 16. CR 403 travels east through the northeast part of Sardinia for 2.55 miles (4.10 km) before coming to an end at Currier–Sardinia Road (CR 404) near the Erie–Wyoming county line.

CR 404, named Currier–Sardinia Road, begins at Olean Road (NY 16) and travels east a fairly short distance to the Wyoming County line. Here, CR 404 ends along with the road name, and the highway heads into Wyoming County as Chaffee Road (unsigned CR 27).

County Route 406, named Matteson Corners Road, is an east–west route in the northern part of the town of Sardinia. CR 406 begins in the northwest part of Sardinia at Pratham Road (CR 407) and travels due east for 3.5 miles (5.6 km) through the town. Very shortly before its east end, CR 406 turns southeast to end at Savage Road (CR 225).

County Route 407, named Pratham Road, is a north–south route in the town of Sardinia that parallels the Concord–Sardinia town line that lies a half-mile to the west. The route begins at an intersection with Middle Road (CR 410) and begins traveling due north through western Sardinia. CR 407 intersects Genesee Road (CR 409) before coming to an end at Crump Road (CR 408) at the Sardinia–Colden town line. Pratham Road itself continues south past CR 410 to an end at Springville–Sardinia Highway (NY 39).

County Route 408 is an overall east–west route in the town of Colden. CR 408, named Crump Road, begins at Center Street (CR 33) and travels east along the Sardinia–Colden town line. The roadway eventually turns northeastward and remains on this alignment to its end at Holland–Glenwood Road (CR 240) near the Colden–Holland town line. Crump Road itself continues west past Center Street another 1.5 miles (2.4 km) to end at Glenwood–East Concord Road (NY 240) near the Kissing Bridge Ski Resort.

County Route 409

County Route 409, named Genesee Road, is an east–west route in the towns of Sardinia and Concord. CR 409 begins at an intersection in the hamlet of East Concord with NY 240 (county-maintained as CR 198 to the south and CR 27 to the north), CR 432, and Genesee Road's westward continuation as CR 49. CR 409 travels west through the south central part of town, and intersects north–south NY 16 in the hamlet of Chaffee. CR 409 continues east to the Wyoming County line where the route ends while Genesee Road continues on.

Genesee Road is 32 miles (51 km) long and stretches from Quaker Street (CR 498) in North Collins to NY 39 in the town of Eagle in Wyoming County. The portion of the highway in Erie County consists of three county routes: CR 73 (7.7 miles or 12.4 kilometres) from CR 498 to the intersection of Wyandale (CR 50) and Mortons Corners–Wyandale Roads (CR 106), CR 49 (6.4 miles or 10.3 kilometres) from there to NY 240, and CR 409.

County Route 410 is an east–west route named Middle Road that runs through the towns of Concord and Sardinia. CR 410 begins at an intersection with CR 228 (Springville–Boston Road). As CR 410 travels east through the southeast portion of Concord, just north of Springville, it intersects NY 240 (Vaughan Street). This section of NY 240 is county-maintained as CR 198. Middle Road continues eastward, entering the town of Sardinia and keeping an eastward track until its end at NY 39 (Springville–Sardinia Highway) just southwest of the hamlet of Sardinia. The roadway named Middle Road continues westward past CR 228 to an intersection with Transit Line Road; however, that section is maintained locally.

It travels north-northwest from this point to an end at CR 228 (Springville–Boston Road), the former routing of US 219 prior to the construction of the Southern Expressway.

The route begins at a cul-de-sac at the foot of US 219 and travels east for 0.4 miles (0.64 km) to an intersection with Transit Line Road (CR 418). Prior to the construction of the Southern Expressway, this roadway used to continue west as Concord Road. It was truncated as a result of the expressway's construction.

At an intersection with Morton Corners–Wyandale Road (CR 106), the route changes names to Concord Road. CR 415 comes to an end at a cul-de-sac at the foot of US 219 (Southern Expressway). Prior to the construction of the Southern Expressway, the roadway continued further east another approximately 1/2 mile to an end at Transit Line Road (CR 418).
