World No. 1 Ashleigh Barty is the defending champion, but she chose not to participate due to safety concerns resulting from the ongoing COVID-19 pandemic.^[1]^[2] This will mark the first time since 2008 that the defending French Open champion would not be participating. This would also be the second consecutive Grand Slam tournament where the defending champion would not be playing (after Bianca Andreescu's withdrawal from the 2020 US Open).^[3]

Simona Halep is in contention for the WTA no. 1 singles ranking. In order to surpass Barty, Halep must win the title.

Serena Williams will attempt to add to her Open era record of 23 Grand Slam singles titles. She will also attempt to equal Margaret Court's all-time record of 24 Grand Slam singles titles.

Angelique Kerber will attempt to become the 11th woman in history to complete a Career Grand Slam in singles.
