The Men's Singles event at the 2010 saw Rafael Nadal, the World No. 1 and 2nd seed, defeat 12th seed Tomáš Berdych 6–3 7–5 6–4 in the final to win his 2nd Wimbledon title, and his 8th Grand Slam title overall.

Roger Federer was the defending champion but Berdych knocked him out in the quarterfinals en route to his final appearance. This was the first time since 2002 that Federer did not make the Wimbledon men's singles final, and the second successive Grand Slam tournament that Federer was defeated at the quarter-final stage, having previously been on a record run of 23 successive Grand Slam semi-finals (of which he won all but three).

For the first time ever at Wimbledon, there were no English players in the men's singles competition, although Great Britain was represented by two players from Scotland: fourth seed Andy Murray and wildcard Jamie Baker.

The first round match between John Isner and Nicolas Mahut set a new record for the longest tennis match in history (both, time and total of games). Isner won the match, taking the final set 70–68 after a total of 11 hours and 5 minutes play across three days.
